First boot tips

I'll be turning on my build for the first time today and I'm looking for some advice on the first boot.

Ill be loading windows 7 from a flash drive on an asus MOBO later I'll post eactly which one but I'm nowhere near it right now.

Really what I need to know is what should the OS file look like for me to install it, right now its a .iso file. should this work?

If need be i could always burn it to a dvd but I figured it would be much faster from a flash.

What usually works best for me is using a piece of software made by microsoft. I think it's called Windows 7 USB tool or something. You then direct it to the ISO file and it does all the rest.
